Use of the semipermeable membrane device as an in situ sampler of waterborne bioavailable PCDD and PCDF residues at sub-parts-per-quadrillion concentrations

Semipermeable membrane devices (SPMDs) were used to passively sample aqueous polychlorinated dibenzo-p-dioxins (PCDDs) and polychlorinated dibenzofurans (PCDFs) in Bayou Meto, AR. The two sites were upstream and downstream from the confluence with a tributary that delivers PCDDs and PCDFs to the Bayou. Following dialysis, cleanup, and fractionation, four replicate 17-9 SPMD samples from each site were analyzed by GUMS, and four were evaluated by H411E bioassay. Traces of only OCDD and HpCDDs were detected in samples from the upstream site. The four samples from below the tributary contained averages of 1550 ± 80 pg of 2,3,7,8-TCDD, 1640 ± 80 pg of 2,3,7,8-TCDF, and lesser quantities of other congeners. The TCDD equivalents obtained by bioassay of replicate SPMD samples agreed well with results obtained by GC/MS. The quantities of 2,3,7,8- TCDD and 2,3,7,8-TCDF sequestered by SPMDs at the downstream site were used to estimate the aqueous concentrations for both compounds as 2 pg/L.

Polychlorinated biphenyls and 2,3,7,8-tetrachlorodibenzo-p-dioxin equivalents in tree swallows from the upper Hudson River, New York State, USA

The upper Hudson River of New York State, USA, is contaminated with polychlorinated biphenyls (PCBs) as a result of industrial discharges throughout the latter half of this century. In 1994 and 1995, we monitored the transfer of PCBs from aquatic sediments to a terrestrial wildlife community using the tree swallow ( Tachycineta bicolor ) as a model organism. Tree swallow eggs and nestlings were collected at four colonies established along a 40-km stretch of the upper Hudson River watershed. Samples were analyzed for total PCBs and PCB congeners, including non- ortho - and mono-ortho-substituted PCBs. Mean concentrations of PCBs in tree swallow eggs and nestlings ranged from 721 to 62,200 ng/g and were as much as 15 times greater than PCB concentrations in tree swallow eggs and nestlings collected from PCB-contaminated areas within the Great Lakes ecosystem. The corresponding 2,3,7,8-tetrachlorodibenzo-p-dioxin equivalents (TEQs) calculated using avian toxic equivalency factors ranged from 410 to 25,400 pg/g. Concentrations of PCB congener 77 (3,39,4,49-tetrachlorobiphenyl) were extremely elevated and were major contributors to the calculated TEQs. Homologue pattern comparisons between Hudson River and Saginaw River (Michigan, USA) ecosystems supported the hypothesis that a consistent Hudson River PCB source was the major contributor to PCBs in Hudson River tree swallows. The high concentrations of PCBs in Hudson River sediments and resultant concentrations observed in tree swallows were indicative of a potential elevated risk to these and other wildlife linked to the aquatic food web of the Hudson River ecosystem.

Investigation of the distribution of organochlorine and polycyclic aromatic hydrocarbon compounds in the Lower Columbia River using semipermeable-membrane devices

Organochlorine and polycyclic aromatic hydrocarbon compounds are of concern in the Columbia River Basin because of their adverse effects on fish and wildlife. Because these compounds can have important biological consequences at concentrations well below the detection limits associated with conventional water-sampling techniques, we used semipermeable membrane devices (SPMDs) to sample water, and achieved sub-parts-per-quintillion detection limits. We deployed SPMDs during 1997 low-flow conditions and 1998 high-flow conditions at nine main-stem sites and seven tributary sites, spanning approximately 700 miles of the Columbia River. We also collected streambed sediment from three sites. SPMD extracts and sediments were analyzed for polychlorinated dibenzo-p-dioxins, polychlorinated dibenzofurans, polychlorinated biphenyls, organochlorine pesticides and related transformation products, and polycyclic aromatic hydrocarbons. Our data indicate that (1) in the absence of additional sources, mechanisms such as volatilization, dilution, and settling of suspended particles can act to significantly reduce concentrations of contaminants along the river's flow path, (2) elevated concentrations of contaminants in the Portland-Vancouver area are primarily from local rather than upstream sources, (3) elevated concentrations of many compounds tend to be diluted during periods of high discharge, (4) much higher discharge in the main stem considerably dilutes elevated concentrations entering from tributaries, (5) the distribution of hydrophobic organic compounds in streambed sediment is not necessarily indicative of their distribution in the dissolved-phase, and (6) SPMDs can reveal patterns of contaminant occurrence at environmentally relevant concentrations that are undetectable by conventional water-sampling techniques.

An automated HPLC method for the fractionation of polychlorinated biphenyls, polychlorinated dibenzo-p-dioxins, and polychlorinated dibenzofurans in fish tissue on a porous graphitic carbon column

The Ah (aryl-hydrocarbon) hydroxylase-receptor active polychlorinated biphenyls (PCBs), polychlorinated dibenzo- p -dioxins (PCDDs) and polychlorinated dibenzofurans (PCDFs) were fractionated by an automated high-performance liquid chromatography (HPLC) system using the Hypercarb™ porous graphitic carbon (PGC) column. This commercially available column was used to fractionate the di-, mono-, and non- ortho PCBs into three fractions for gas chromatography (GC)/electron capture detection analysis, and a fourth fraction containing the PCDDs/PCDFs for GC/mass spectrometry analysis. The recoveries of the PCBs ranged from 68 to 96%, and recoveries of the PCDDs/PCDFs ranged from 74 to 123%. The PGC column has the advantage of faster separations (110 min versus 446 min) and less solvent use (275 ml versus 1,100 ml) compared with automated fractionation of these compounds on activated carbon (PX-21), while still affording good separation of the classes. The PGC column may have an advantage over the pyrenyl-based HPLC method because it has a greater loading capacity (400 μg total PCBs versus 250 μg). Overall, the PGC is a standard column that provides reproducible fractionation of PCDD/PCDFs and PCBs for analytical measurement in environmental samples.

Polychlorinated dibenzo-p-dioxins, dibenzofurans, biphenyls and 2,3,7,8-tetrachlorodibenzo-p-dioxin equivalents in fishes from Saginaw Bay, Michigan

Concentrations of polychlorinated biphenyls (PCBs), polychlorinated dibenzo- p -dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs), polychlorinated naphthalenes (PCNs), polychlorinated dibenzothiophenes (PCDTs), and 2,3,7,8-tetrachlo-rodibenzo- p -dioxin (2,3,7,8-TCDD) equivalents were measured in composite samples of forage fishes and several sizes of walleyes ( Stizostedion vitreum ) and common carp ( Cyprinus carpio ) collected from Saginaw Bay, Michigan, USA. The concentrations of 2,3,7,8-TCDD equivalents determined from the use of TCDD equivalency factors (TEFs) and instrumentally determined concentrations of individual congeners were designated TEq. The 2,3,7,8-TCDD equivalents determined on the same extracts with the H4IIE bioassay were defined as TCDD-EQ. Concentrations of TCDD-EQ and TEq were significantly, but poorly correlated ( R 2 = 0.44). Concentrations of TCDD-EQ and TEq ranged from 10.6 to 348 and 13.5 to 69.5 ng/kg, wet weight, respectively. The ratio of TCDD-EQ to TEq ranged from 0.5 to 5.0, with the greatest ratio observed in common carp but the average ratio between TEq and TCDD-EQ was approximately 1.0. In common carp, larger walleye, and medium-sized alewife ( Alosa pseudoharengus ) there were significant concentrations of TCDD-EQ that could not be accounted for by TEq calculated from an additive model of PCBs, PCDDs, or PCDFs. As much as 75% of the TCDD-EQ in some samples could not be accounted for by TEqs. It is unlikely that PCNs, PCDTs, or polychlorinated diphenyl ethers (PCDEs) occurred at concentrations that were, based on their relative potencies, great enough to contribute significant quantities of TCDD-EQ. The discrepency between TCDD-EQs and TEqs was not due to antagonisms among congeners or extraction efficiencies.

Dietary exposure of mink to carp from Saginaw Bay. 3. Characterization of dietary exposure to planar halogenated hydrocarbons, dioxin equivalents, and biomagnification

Mink are known to be very sensitive to the toxic effects of planar polychlorinated biphenyls (pPCBs), polychlorinated dibenzo- p -dioxins (PCDDs), and polychlorinated dibenzofurans (PCDFs), collectively known as planar halogenated hydrocarbons (PHHs). Previously, we reported the reproductive effects in mink fed a diet containing 10, 20, or 40% fish taken from Saginaw Bay, Lake Huron. The present study reports the chemical characterization of the diets and the adult mink livers, along with a comparison of an additive model of toxicity with the results of the H4IIE bioassay on these samples. The assessment of dietary or tissue-based exposure of the mink to 2,3,7,8-tetrachlorodibenzo- p -dioxin (TCDD) and related compounds revealed that TCDD equivalents of the PHH mixtures largely followed an additive model of toxicity as compared with the H4IIE bioassay. Consistent dietary and liver tissue-based threshold concentrations for reproductive toxicity in mink were determined regardless of whether PHHs were quantified as TEQs (additive toxicity) or TCDD-EQs (H4IIE bioassay). Significant reproductive effects were observed in the lowest treatment group (10% fish or 19.4 pg of H4IIE bioassay-derived TCDD-EQs/g). Consumption-normalized mink liver biomagnification factors (BMFs) were 6.4−74.2 for PCDDs, <1−75.8 for PCDFs, <1−15.9 for PCBs, and in general, increased with degree of chlorination within each class. Based on TEQs or TCDD-EQ, this study confirms that mink are among the most, if not the most, sensitive mammalian species to the reproductive toxicity of TCDD and related compounds.
